OPPORTUNITY
Mammoth Biosciences is seeking a Senior Scientist I for its Protein Engineering team. This role will execute design-build-test-learn cycles to create and optimize proteins for gene editing applications.The successful candidate will be responsible for proposing and executing strategies for high-throughput generation and screening of proteins involved in CRISPR systems for gene editing applications. This role will function as part of the Protein Engineering group, which is responsible for optimizing and creating proteins across all gene editing modalities being pursued at Mammoth Biosciences.

KEY RESPONSIBILITIES- High-throughput molecular cloning and diversity generation for protein engineering targets
- High-throughput screening in mammalian systems for increased potency and specificity in gene editing outcomes
- Design and use of both pooled and arrayed screening strategies for efficient identification of efficacious variants and fusion proteins
- Use of NGS and FACS assays for ascertaining editing outcomes
- Identification of structure-activity relationships from engineering outcomes
- Nomination of key molecules for downstream testing by project teams
- Working with editing technology project teams to optimize new gene editing modalities
- Analysis of large datasets
- Documentation of experimental results in laboratory notebooks, present experimental results at internal meetings, and other duties as assigned
REQUIRED QUALIFICATIONS- Bachelors, Masters, or Ph.D. in Biochemistry, Bioengineering, Molecular Biology, or related area
- Postgraduate experience in years (industry/postdoc): Bachelor's 8+, Masters 6+, or PhD 3+
- Expertise in high-throughput methods for protein diversity generation
- Expertise in arrayed and pooled (e.g. deep mutational scanning) screening approaches in mammalian cells with NGS and FACS-based readouts
- Experience in constructing reporter cell lines for FACS-based screens
- Experience with analysis of large datasets
PREFERRED QUALIFICATIONS- Familiarity with liquid handlers such as Hamilton/Tecan/ECHO
- Familiarity with colony pickers such as Qpix
- Experience with CRISPR systems and/or other gene editing technologies
- Experience leading and mentoring scientists and collaborating cross functionally
- Computer and scripting skills (Benchling, Geneious, JMP, Python, FlowJo)

Base Salary Range: $ 155,000 - $165,162
The listed base salary range is for Mammoth employees in the Bay Area. Actual base salary will be determined by geographic work location, relevant professional experience, applicable skills, and internal equity. The base salary range for those working remotely outside of the San Francisco Bay Area may differ and will be determined by industry market data for the role and specific region.
